Vikram Banerjee claimed four wickets, but could not prevent Derbyshire gaining a first innings lead of 103 on the second day at Derby.
The home side began the day on 71-3 and centuries from Chris Rogers and 19-year-old Chesney Hughes, his first in the Championship, helped them to 345 all out.
Banerjee took 4-62 from 27 overs, while Gemaal Hussain returned 3-65 from 18. By the close Gloucestershire had reached 47-1 in their second innings, losing Steve Snell for 21.
Opener Rogers was unbeaten on 49 overnight and the second ball of the morning saw him reach his half-century off 75 balls, with eight fours.
It took him only another 60 deliveries to reach his fourth hundred of the season. He took four boundaries off one Alex Gidman over and then struck Banerjee to mid-wicket for four to bring up the 45th first class century of his career.
The Derbyshire skipper was eventually caught behind for 115 shortly before Hughes passed 50 in 109 deliveries with 7 fours. Lunch was taken at 195-4.
A first innings lead for the home side still looked in doubt as Dan Redfern (12), Robin Peterson (8), Tom Poynton (22) and Tom Lungley (1) all fell to the left-arm spin of Banerjee.
Hughes stood firm, however, and struck consecutive boundaries to get to his hundred shortly before tea, having faced 196 balls and hit 14 fours. By the interval Derbyshire were 285-8 and already 43 in front.
The aggressive Hughes launched Banerjee over mid-on for six early in the evening session and Tim Groenewald offered valuable support in a ninth-wicket partnership of 43.
Hughes never looked in danger until Groenewald’s straight drive was palmed onto the stumps at the bowler's end by Jon Lewis, leaving the Derbyshire hero short of his ground, having made 118.
Groenewald (34) was then supported by Atif Sheikh as a further 29 runs were added for the final wicket before the number 11 edged Lewis to Snell, who had replaced the injured Jonathan Batty behind the stumps, to bring the innings to an end.
Ten overs remained in the day and Greg Smith grabbed the wicket of Snell, clean bowled, with three overs left. Nightwatchman Steve Kirby helped Abdul Kadeer-Ali (21 not out) see Gloucestershire to the close.
