A career-best 172 from Ben Wright and a century by Jim Allenby made it a day of toil for Gloucestershire's bowlers at the Swalec Stadium.
But there was still a five-wicket haul for Anthony Ireland as Glamorgan extended their first innings score from an overnight 257-3 to 583-9 declared.
Ireland returned 5-114 from 29.5 overs, having taken seven wickets in the previous Championship game against Leicestershire, his first four-day appearance of the season.
But in all other respects it was Glamorgan's day. Wright, unbeaten on 77 at the start, ended up blasting 23 fours and 3 sixes in a brilliant 219-ball innings he will long remember.
The 22-year-old former England Under-19 batsman dominated a fourth-wicket stand of 157 with Mike Powell (37) which took Glamorgan's total to 360 before Powell was bowled by James Franklin.
The pair had batted right through the morning session, Wright completing his second Championship hundred from 140 balls.
He put Gloucestershire's attack to the sword before perishing with the total on 390, edging Jon Lewis to wicketkeeper Jonathan Batty having batted for over four hours.
Mark Wallace failed to trouble the scorers before being caught by Chris Dent off Ireland, but it was a rare blemish in an impressive Glamorgan batting performance.
Allenby had come in at the fall of Powell's wicket and soon took over where Wright had left off, guiding the home side to maximum batting points as he reached a century off 167 deliveries, with 8 fours and a six.
Robert Croft hit 6 fours in reaching 63 off 116 balls and the scoreboard read 545-7 by the time he was stumped by Batty advancing to Vikram Banerjee.
Allenby finally followed on 564, caught by Franklin off Ireland, who then claimed his fifth victim when David Harrison was picked up by Dent for ten.
That brought the declaration with Glamorgan 166 runs ahead. leaving Gloucestershire an awkward eight overs to negotiate before the close.
They didn't quite manage it as Dent shouldered arms to Dean Cosker's final ball of the day and was bowled for three. The young batsman left the pitch disconsolate with his team 12-1 and facing a tough final day.
